中文摘要 沙門氏菌(Salmonella)為引起細菌性食物中毒的細菌之一,且其食物中毒致死率為最高,因此發展靈敏且快速檢測病原菌的技術為重要公衛議題。本計畫提出整合免疫奈米磁珠(magnetic nanoparticles, MNPs) 於Salmonella濃縮、分離與檢測的免標定式阻抗感測技術。以修飾有7F1A抗體(為Ab1) 的MNP(Ab1@MNPs)對Salmonella先進行免疫分離與濃縮,以提高菌數濃度與去除背景干擾物影響。應用交流電動流(alternating current electrokinetic flow, ACEKF)傳輸含有Salmonella/Ab1@MNP菌液與利用介電泳 (dielectrophoresis, DEP) 力濃縮該粒子於檢測電極表面,可提升電極表面的免疫反應效率,縮短免疫反應時間,降低可測得之細菌密度,提升檢測靈敏度。本計畫所開發之可快速檢測食品微生物之免標定式阻抗型免疫檢測平台不僅能用於食品安全領域,且能應用於其他免疫分析。
英文摘要 Salmonella, being one of bacteria causing foodborne illness, has the highest mortality in food poisoning. Therefore, developing a sensitive and rapid method for detecting bacteria is an important issue in the sanitary field. This study proposes a label-free impedimetric biosensing technique with immune magnetic nanoparticles (MNPs) for enrichment, separation and detection of Salmonella. The MNPs modified 7F1A antibody (Ab1) reacts with Salmonella for separation and enrichment, which can increase the concentration of bacteria and prevent the effect of interference. Salmonella/Ab1@MNPs complexes are transported to the detector region by alternating current electrokinetic flow (ACEKF) and are concentrated to the surface of detector by dielectrophoresis (DEP), which can increase immunoreaction rate, shorten response time, lower the detection limit of bacteria and improve sensitivity. The label-free impedimetric immunosensing platform of this study for the detection of foodborne bacteria can be used not only in food safety field but also in other immunoassay applications.
